Heterogeneity of human platelets. VI. Correlation of platelet function with platelet volume.

چکیده
Platelet aggregation velocity was moosured with an aggregometer in plateletrich plasma from 34 normal subjects following addition of ADP, collagen, or epinephrine. Platelet volume parameters [mean, mode, megathrombocyte index (large platelets)] were determined with a model B Coulter Counter attached to a P64 Channel Analyzer with electronic recorder and correlated with platelet count. A negative linear relationship was found between platelet count and platelet volume (r = -0.53, p < 0.001). Platelet aggregation velocity induced with ADP, collagen, or epinephrine ( primary wave) was directly proportional to platelet volume and correlated best with megathrombocyte index: ADP, r = 0.62, p < 0.001; collagen, r = 0.59, p < 0.001; epinephnne, r = 0.53, p < 0.02. Platelet aggregation velocity induced with collagen or epinephrine correlated poorly with platelet moon or mode. Platelet aggregation velocity induced with ADP did not correlate with p1 atelet moon or mode volume (r = 0.27, p>O.l; r0.22, p>O.l, respectively). Platelet volume distribution curves of residual nonaggregated platelets revealed a relative absence of larger platelets. Thus, platelet aggregation velocity was propertional to platelet volume, but correlated best with the number of large platelets or megathrembocyte index, particularly with ADP-induced platelet aggregation. It is suggested that the clinical strategy for treatment of platelet disorders would be better directed toward evaluation of platelet size and function rather than number, and that experimental as well as therapeutic procedures for the harvesting of platelets be directed toward a method of collection in which a portion of the megathrombocyte population is not routinely discarded.

Synergic effects of Summer Savory extract and aspirin on human platelets function
Introduction: Cardiovascular disease is one of the most common diseases in the world. In this account, atherosclerosis is one of the main cause’s heart attacks. Coagulation activity of platelets plays an important role in atherosclerotic plaque formation, thrombosis formation and cardiovascular diseases. Aspirin is the most commonly drugs which used as anti-platelet function to prevent and trea...
متن کاملPlatelet Factor 3 Based-clotting Time Assay as a Quality Marker for Long-term Storage of Platelet Concentrates
Background: Platelets rapidly lose their qualities usually after 5 day of storage. Different standard methods have been recommended to check the quality of platelets during storage which some of them show better correlation with other quality markers during storage. The purpose of this study was to demonstrate if platelet factor 3 (PF3) assay could be an indicator of storage lesion and provide ...

Background: Coagulation activity of platelets plays an important role in thrombosis formation, Atherosclerotic plaque establishment and cardiovascular disorders. ASA are using as an anti platelet activity drug for patients with Coronary Artery Disease (CAD). Using of plant extraction as a complementary medicine for these patients (CAD) is mostly recommended by traditional medicine specialist. ...

Background: Major platelet adhesive receptors that contribute significantly to thrombus formation include platelet receptor glycoprotein Ibα (GPIbα) of the GPIb-IX-V complex and platelet glycoprotein VI (GPVI). GPIbα plays a crucial role in platelet tethering to sub-endothelial matrix, which initiates thrombus formation at arterial shear rates, whereas GPVI is critically involved in platelets f...
متن کاملEvaluation of microRNAs; mir223, mir222 and mir92a levels in the Platelet-derived microparticles in the Platelet concentrates produced by Platelet Rich Plasma method during storage
Abstract Background and Objectives Platelets release microparticles containing cellular components, including microRNAs, during storage. Assessment of these microRNAs is one of the markers for evaluation of platelet storage lesions. The aim of the present study was to evaluate the level of changes in the expression of mir-223, mir-92a and mir-222 during storage in platelets prepared by platele...
